Abstract: Pepaya dan kunyit putih merupakan tanaman yang memiliki kandungan senyawa aktif yang dapat dimanfaatka sebagai antibakteri. Daun pepaya diketahui mengandung alkaloid, saponin, dan flavonoid. Kunyit putih mengandung flavonoid dan alkaloid. Kandungan senyawa aktif pada daun papaya dan kunyit diharapkan dapat diekstraksi menjadi senyawa antibakteri. Penelitian ini dilaksanakan di Laboratorium Biologi Molekuler Fakultas Kedokteran Universitas Prima Indonesia. Metode yang digunakan dalam penelitian ini adalah eksperimental laboratorium dengan rancangan acak lengkap. Data yang diperoleh diolah menggunakan software SPSS versi 23 dan analisis lanjut dengan DNMRT. Penelitian terdiri dari (sebutkan berapa tahap) yaitu: Variasi konsentrasi ekstrak yang digunakan adalah 25%, 50%, 75% dan 100%. Kontrol positif pengujian menggunakan klorampenikol (2 ug). Berdasarkan hasil skrining fitokimia ekstrak daun pepaya mengandung flavonoid, alkaloid, terpenoid, dan saponin. Sedangkan ekstrak kunyit putih mengandung flavonoid, steroid, tanin dan saponin. Hasil uji antagonis menunjukkan konsentrasi ekstrak 100% menghasilkan nilai zona hambat terbesar. Nilai zona hambat yang dihasilkan oleh ekstrak papaya 100% terhadap E.coli dan S. epidermidis masing-masing sebesar 14,110 mm dan 14,707 mm. Nilai zona hambat yang dihasilkan oleh ekstrak kunyit putih 100% terhadap E.coli dan S. epidermidis masing- masing sebesar 14,307 mm dan 13,743 mm. The aim of this research is; To determine the active compounds contained in papaya leaf extract (Carica papaya) and white turmeric extract (Curcuma amada), To determine the antimicrobial activity of papaya leaf extract (Carica papaya) and white turmeric extract (Curcuma amada) against Escherichia coli and Staphylococcus epidermidis, To determine the minimum inhibitory concentration of papaya leaf extract (Carica papaya) and white turmeric extract (Curcuma amada) against Escherichia coli and Staphylococcus epidermidis. This research was carried out from September to November 2020, at the Laboratory of Molecular Biology, Faculty of Medicine, University of Prima Indonesia. The method used in this research was experimental laboratory with a completely randomized design. The treatment concentration in 2 extracts such as 25%, 50%, 75% and 100% and the positive control used was chloramphenicol (2 ug). The data obtained were processed using SPSS version 23 software and further analysis with DNMRT. Based on the results of the study, the results of phytochemical screening of papaya leaf extract contained potential secondary metabolites such as flavonoids, alkaloids, terpenoids, and saponins. Furthermore, in white turmeric extract there are flavonoids, steroids, tannins and saponins. Giving 100% papaya leaf extract is the best treatment in suppressing the growth of Echerichia coli and Staphylococcus epidermidis bacteria with an average inhibitory power of 14.110 mm and 14.707 mm. Giving 100% white turmeric extract is the best treatment in suppressing the growth of E.coli and S. epidermidis with an average inhibition of 14,307 mm and 13,743 mm.
