The quantity and quality of water are absolutely considered because water is a basic human need. Bacterial contamination in water can be harmful to health. This study aims to determine whether there is contamination of Coliform in water in 10 villages in Serdang Bedagai Regency, in refill drinking water at a depot using raw water from groundwater, to compare the effectiveness of the two series of ultrafiltration methods in separating Coliform from refill drinking water, to determine the characterization of Coliform after the ultrafiltration process. The research method consisted of field observations, random sampling and identified quantitatively and qualitatively to see the characterization of Coliform, then the results of the analysis are described descriptively. The results of the study on ten groundwaters showed that all of them were identified as containing Coliform, and one village, namely Tambak Cekur village, had groundwater that met the requirements. Ultrafiltration method is still less effective in reducing Coliform content because two out of three samples of drinking water were still identified as containing Coliform. The comparison of the two series of ultrafiltration methods shows that the second ultrafiltration method (Filter + Carbon Block + GAC) is more effective in reducing Coliform content than the first ultrafiltration method (Filter). Coliform bacteria identified after the ultrafiltration process were Enterobacter cloacae, and Enterobacter aerogenesen_US
