Abstract: | Salah satu patogen utama yang menyebabkan penurunan produksi tanaman cabai ialah Pepper Yellow Leaf Curl Virus (PepYLCV). Penyakit kuning keriting ini disebabkan oleh begomovirus (genus: Geminivirus) yang ditularkan melalui serangga vektor kutu kebul (B. tabaci). Salah satu komponen gen pengkode protein fungsional yang berperan penting dalam penyebaran partikel virus adalah gen Rep (C1), Gen Rep (C1) adalah elemen pengatur replikasi dan transkripsi pada geminivirus. Adanya infeksi dari virus PepYLCV ini mengakibatkan penurunan produksi tanaman cabai di Kabupaten Karo, Sumatera Utara. Oleh karena itu Tujuan penelitian ini adalah untuk untuk menganalisa DNA Virus PepYLCV yang sudah terintegrasi oleh PCR dengan menggunakan primer spesifik C1-TD21BamHi-NT dan C1-TD22Smal-N ke dalam genom tanaman cabai di Kabupaten Karo, Sumatera Utara. Hasil penelitian menunjukkan PepYLCV pada sampel Lokasi 3 dengan panjang 1070 Bp. Isolat sampel lokasi 3 memiliki keragaman molekuler dengan isolat kode (LC547427.1), (NC008283.1) dan (AB267834.1), dengan tingkat kemiripan masing- masing 97,71%, 96,37% dan 96,37%. One of the main pathogens that causes a decrease in chili plant production is Pepper Yellow Leaf Curl Virus (PepYLCV). Curly jaundice is caused by begomovirus (genus: Geminivirus) which is transmitted via the whitefly vector insect (B. tabaci). One of the functional protein coding gene components that plays an important role in the spread of viral particles is the Rep gene (C1). The Rep gene (C1) is a replication and transcription regulatory element in geminiviruses. The presence of infection with the PepYLCV virus has resulted in a decrease in chili plant production in Karo Regency, North Sumatra. Therefore, the aim of this research is to analyze PepYLCV Virus DNA which has been integrated by PCR using specific primers C1-TD21BamHi-NT and C1-TD22Smal-N into the chili plant genome in Karo Regency, North Sumatra. The results showed that PepYLCV in the Location 3 sample was 1070 Bp long. The isolate from location 3 samples has molecular diversity with isolates coded (LC547427.1), (NC008283.1) and (AB267834.1), with similarity levels of 97.71%, 96.37% and 96.37%, respectively. |
