Abstract: Kawasan Air Terjun Sibolangit di Sumatra Utara kaya akan keanekaragaman hayati, termasuk lumut hati bertalus Marchantia emarginata yang berpotensi sebagai sumber senyawa bioaktif. Penelitian ini bertujuan mengidentifikasi bakteri endofit pada lumut hati bertalus dan menguji aktivitas antibakterinya terhadap Escherichia coli dan Staphylococcus aureus. Penelitian ini merupakan penelitian deskriptif yang dilakukan dalam dua tahap. Tahap pertama adalah pengambilan sampel lumut hati dari lokasi Air Terjun Satu Hati di Kecamatan Sibolangit menggunakan metode survei eksploratif. Tahap kedua adalah identifikasi bakteri endofit dan pengujian aktivitas antibakteri menggunakan metode difusi cakram yang dilaksanakan di Laboratorium Universitas Sari Mutiara Indonesia, Kota Medan. Data yang diperoleh dianalisis secara kualitatif dan kuantitatif. Hasil penelitian menemukan lima isolat bakteri endofit dari lumut hati yang menunjukkan aktivitas antibakteri. Isolat SP5 memberikan zona hambat terbesar terhadap E. coli (20,89 mm, sangat kuat), sementara isolat SP1 menunjukkan zona hambat 19,38 mm terhadap S. aureus (sedang). Berdasarkan hasil tersebut dapat disimpulkan bakteri endofit dari lumut hati memiliki potensi sebagai agen antibakteri terhadap kedua bakteri patogen tersebut. The Sibolangit Waterfall area in North Sumatra is rich in biodiversity, including the thallus liverwort Marchantia emarginata which has the potential as a source of bioactive compounds. This study aims to identify endophytic bacteria in thallus liverwort and test its antibacterial activity against Escherichia coli and Staphylococcus aureus. This study is a descriptive study conducted in two stages. The first stage is the sampling of liverwort from the Satu Hati Waterfall location in Sibolangit District using an exploratory survey method. The second stage is the identification of endophytic bacteria and testing of antibacterial activity using the disc diffusion method carried out at the Laboratory of Sari Mutiara Indonesia University, Medan City. The data obtained were analyzed qualitatively and quantitatively. The results of the study found five isolates of endophytic bacteria from liverwort that showed antibacterial activity. Isolate 𝑆𝑃5 gave the largest inhibition zone against E. coli (20.89 mm, very strong), while isolate 𝑆𝑃1 showed an inhibition zone of 19.38 mm against S. aureus (moderate). Based on these results, it can be concluded that endophytic bacteria from liverwort have the potential as antibacterial agents against both pathogenic bacteria.
