Abstract: | Penelitian ini bertujuan untuk mengetahui genus bakteri endofit asal tanaman kopi dan sebagai agen biokontrol terhadap penyakit busuk buah (Fusarium sp.). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) non faktorial yang terdiri dari 7 perlakuan dengan 3 ulangan. Perlakuan yang diuji terdiri dari : P0 = Kontrol negatif, P1 = Kontrol Positif (penggunaan fungisida sintetik dengan bahan aktif Benlate), P2 = Bakteri Endofit Isolat 01, P3 = Bakteri Endofit Isolat 02, P4 = Bakteri Endofit Isolat 03, P5 = Bakteri Endofit Isolat 04, P6 = Bakteri Endofit 05. Dari Isolasi bakteri endofit tanaman kopi diperoleh 6 isolat bakteri endofit. Karakterisasi morfologi melibatkan pengamatan bentuk, margin,elevasi, dan warna. Hasil uji pH menunjukkan bahwa semua isolat memiliki pH sekitar 6,5 yang merupakan kondisi netral untuk pertumbuhan bakteri. Isolat bakteri endofit menunjukkan aktivitas antimikroba terhadap mikroba patogen, terlihat dari zona hambat yang terbentuk. Didapatkan isolat bakteri endofit yang berpotensi sebagai agen biokontrol pada tanaman kopi pada kode isolat BETK 05 pada pengamatan 2 x 24 jam dengan rataan 9,40mm dan BETK 04 dengan rataan 8,36mm menunjukkan zona hambat paling besar. From the i.volation of endophytic bacteria from coffee plants, 6 isolates of endophytic bacteria were obtained. Morpholngica/ cltarae-·teri:micm involved ohsen,alion of shape, margin, elevation, and color. pH test results showed that all i~'Oiales had a pH of around 6.5, which was a neutral condition for bacterial growth. The endophytic bacterial isolates showed antimicrobial activity against pathogenic microbes, as seen from the inhibition zones formed. Endophytic bacterial isolates with potential as biocontrol agents in coffee plants were obtained in isolate code BETK 05 with an average inhibition zone of9.40 mm and BETK 04 with an average of8.36 mm al 2 x 2-1 how·s observation, showing the largest inhibition zones. |
