Abstract: | Penelitian ini bertujuan untuk mengkaji dan memberikan informasi mengenai perkembangan dan virulensi Metarhizium anisopliae pada beberapa media cair terhadap larva kumbang tanduk Oryctes rhinoceros (Coleoptera: Scarabaeidae). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) non faktorial, dengan 4 perlakuan dan 5 ulangan sehingga terdapat 20 unit percobaan, terdiri dari: H1 = Media Potato Dextrose Broth (PDB), H2 = Media Sabouroud Dextrose Yeast extract Broth (SDYB), H3 = Media Yeast extract Peptone Glucose Broth (YPGB) dan H4 = Air Kelapa Tua. Data hasil penelitian ditransformasi menggunakan akar kuadrat (SQRT) kemudian dianalisis dengan Analysis of Variance (ANOVA) berdasarkan model linear dari rancangan yang digunakan. Apabila hasil ANOVA menunjukkan hasil berbeda nyata dan sangat nyata, maka dilanjutkan dengan Uji beda rataan menurut Duncan’s Multiple Range Test (DMRT) pada taraf kepercayaan 5%. Hasil penelitian menunjukkan bahwa 1. Pengembangan M. anisopliae pada beberapa media cair mempengaruhi tipe spora M. anisopliae. Spora yang dihasilkan dari perlakuan HI (media Potato Dextrose Broth), H2 (media Sabouroud Dextrose Yeast extract Broth) dan H3 (media Yeast extract Peptone Glucose Broth) merupakan spora konidiospor sedangkan perlakuan H4 (air kelapa tua) menghasilkan spora blastospor dan konidiospor. 2. Respon perlakuan pengembangan M. anisopliae pada beberapa media cair berpengaruh sangat nyata terhadap kerapatan spora, viabilitas spora dan persentase mortalitas larva Oryctes rhinoceros. Perlakuan terbaik terdapat pada perlakuan H4 atau media air kelapa tua dimana persentase mortalitas pada hari ke 12 setelah aplikasi sudah mencapai mortalitas 100% dengan nilai LT50 7,16 hari. This research aims to study and provide information regarding development and virulence Metarhizium anisopliae in several liquid media against horn beetle larvae Oryctes rhinoceros (Coleoptera: Scarabaeidae). This research used a non-factorial Completely Randomized Design (CRD), with 4 treatments and 5 replications so that there were 20 experimental units, consisting of: H1 = Media Potato Dextrose Broth (PDB), H2 = Media Sabouroud Dextrose Yeast extract Broth (SDYB), H3 = Media Yeast extract Peptone Glucose Broth (YPGB) and H4 = Old Coconut Water. The research data was transformed using square roots (SQRT) and then analyzed using Analysis of Variance (ANOVA) based on the linear model of the design used. If the ANOVA results show significantly or very significantly different results, then proceed with the mean difference test according to Duncan’s Multiple Range Test (DMRT) at a 5% confidence level. The results showed that 1. The development of M. anisopliae in several liquid media influenced the type of M. anisopliae spores. The spores produced from the HI (Potato Dextrose Broth media), H2 (Sabouroud Dextrose Yeast extract Broth media) and H3 (Yeast extract Peptone Glucose Broth media) treatments are conidiospores, while the H4 (old coconut water) treatment produces blastospores and conidiospores. 2. The response to the development treatment of M. anisopliae in several liquid media has a very significant effect on spore density, spore viability and the percentage of mortality of Oryctes rhinoceros larvae. The best treatment is in the H4 treatment or old coconut water media where the percentage of mortality on the 12th day after application is already achieved 100% mortality with LT50 of 7,16 days. |
