Abstract: Air merupakan zat yang sangat dibutuhkan oleh makhluk hidup untuk aktivitas kehidupannya. Menurut Permenkes Nomor 2 Tahun 2023, air yang digunakan untuk higiene dan sanitasi harus bebas dari kontaminasi mikrobiologi, fisik dan kimia. Penelitian ini bertujuan untuk menganalisa kualitas fisik dan biologi air sumur gali di Desa Patumbak Kecamatan Patumbak Kabupaten Deli Serdang. Analisis kualitas fisik dilakukan secara organoleptik dan menggunakan digital water tester dan analisis kualitas biologi menggunakan koliform kit. Hasil penelitian menunjukkan bahwa 5 dari 10 sampel yang diuji tidak memenuhi syarat fisik dan pH yaitu pada sampel 1, 3, 6, 9 dan 10. Sampel 2,4,5,7 dan 8 memenuhi syarat yaitu tidak berwarna, tidak berbau, tidak berasa, nilai TDS >300 mg/L dan nilai pH masih dalam rentang 6,5 – 8,5. Analisis kualitas biologi menunjukkan bahwa semua sampel air sumur gali mengandung bakteri Coliform. Sedangkan pada uji Escherichia coli terdapat 8 sampel mengandung bakteri Escherichia coli yaitu pada sampel 1, 2, 4, 5, 6, 7, 9 dan 10. Sampel yang tidak mengandung bakteri Escherichia coli yaitu 3 dan 8. Berdasarkan hasil penelitian dapat disimpulkan bahwa 100% sampel air sumur gali di di Dusun IV Kecamatan Patumbak Kabupaten Deli Serdang tidak memenuhi persyaratan kualitias fisik dan biologi sesuai Permenkes Nomor 2 Tahun 2023. Water is a substance that is needed by living creatures for their life activities. According to Minister of Health Regulation Number 2 of 2023, water used for hygiene and sanitation must be free from microbiological, physical and chemical contamination. The purpuse of this research was to determine the physical and biological quality of dug well water in Patumbak Village, Patumbak District, Deli Serdang Regency. Physical quality analysis was carried out organoleptically and using a digital water tester and biological quality analysis using a coliform kit. The results showed that 5 of the 10 samples tested did not meet the physical and pH requirements, samples 1, 3, 6, 9 and 10. Samples 2, 4, 5, 7 and 8 met the requirements, colorless, odorless, tasteless, the TDS value is >300 mg/L and the pH value is still in the range of 6.5 – 8.5. Biological quality analysis shows that all dug well water samples contain Coliform bacteria. Meanwhile, in the Escherichia coli test, there were 8 samples containing Escherichia coli bacteria, samples 1, 2, 4, 5, 6, 7, 9 and 10. Samples that did not contain Escherichia coli bacteria were 3 and 8. Based on the research results, it can be concluded that 100 % of dug well water samples in IV Village, Patumbak District, Deli Serdang Regency do not meet the physical and biological quality requirements according to Minister of Health Regulation Number 2 of 2023.
